How do I send or reply to email using my new trashmail address in the from field?

For reply, just use reply on your email client. Trashmail changes default reply to pass through trashmail server to protect your privacy.

Yes. Login to your email account at trashmail web site and use the link that gets you there: https://ssl.trashmail.net/?login=1&auth=1&cmd=send_mail

Do I need a paid account to do this?

As far as I know: No.

What client do you use? Website directly, or Firefox Add-On?

Some advanced mail clients have an editable "From:" field, where you can freetype whatever you want. Mutt is one such MUA. With mutt, there is no need to route replies through trashmail.net.
